Findings
This variant was found in a family of the Chinese Familial Alzheimer’s Disease Network in which five family members were affected, spanning two generations (Jia et al., 2020). Mean age at onset was 47 years, ranging between 45 and 49 in the four affected carriers who were genotyped and found to carry the mutation. No unaffected non-carriers were reported to confirm co-segregation. Three of the four L172W carriers had an APOE E4/E3 genotype, and one was homozygous for APOE3.
This variant was absent from the gnomAD variant database (gnomAD v2.1.1, Aug 2021).

Unknown.
Biological Effect
The biological effect of this variant is unknown, but four of five in silico algorithms (Polyphen2, PANTHER, Mutpred2, PROVEAN, Mutation Taster) predicted it has a damaging effect (Jia et al., 2020). The PHRED-CADD score was 22.8.
