Findings
In a European-American cohort of 134 sporadic late-onset Alzheimer’s cases and 266 controls, this variant was found with a minor allele frequency of 5.6×10-3 (three of 532 alleles) among the controls. The F1291L was not found by whole- exome or genome sequencing of 866 familial late-onset AD cases and 324 controls in the same study (Fernández et al., 2016).
In a multi-center European study of 1255 cases and 1938 controls from the European Early-Onset Dementia Consortium, one 57-year-old Italian control subject was found to be a heterozygous carrier of the F1291L variant (Verheijen et al., 2016).
This variant also was reported in one of 5198 AD cases and one of 4491 controls in a dataset from the Alzheimer’s Disease Sequencing Project (ADSP), consisting of subjects of non-Hispanic Caucasian ancestry from whom whole-exome sequencing data were available (Campion et al., 2019).
In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, including ADSP, this allele was observed four times—twice among the AD cases and twice among the controls (Holstege et al., 2022).
The F1291L variant is classified as likely benign by the criteria of Holstege et al. (Holstege et al., 2017).
Functional Consequences
The variant was predicted to be tolerated by SIFT, disease-causing by Mutation Taster, and benign by PolyPhen-2 (Verheijen et al., 2016).
