Findings

In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, this allele was observed once among the AD cases (Holstege et al., 2022).

Previously, a 75-year-old Belgian control subject was reported to be a heterozygous carrier of this variant, in a European sample of 1255 Alzheimer’s cases and 1938 controls (Verheijen et al., 2016).

The variant was classified as “uncertain: possibly pathogenic” by the criteria of Holstege et al., 2017 (Holstege et al., 2017).

Functional Consequences

The L807P variant was predicted to be damaging by SIFT, disease-causing by Mutation Taster, and probably damaging by PolyPhen-2 (Verheijen et al., 2016).
