Findings
In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, this allele was observed nine times—four times among the AD cases and five times among the controls (Holstege et al., 2022). A mega-analysis of these data did not find an association between the variant and AD risk.
Previously, in a Dutch sample of 640 Alzheimer’s cases and 1268 controls, a 53-year-old AD patient was found to be a heterozygous carrier of this variant (Holstege et al., 2017). Two additional carriers were reported in data sets from the Alzheimer’s Disease Sequencing Project (5198 AD cases and 4491 controls of non-Hispanic Caucasian ancestry, from whom whole-exome sequencing data were available) and the Alzheimer Disease Exome Sequencing France (ADESFR) project (927 late-onset Alzheimer’s cases, 852 early-onset cases, and 1273 controls) (Campion et al., 2019). Both of these carriers were classified as early onset cases. ADSP, ADESFR, and the cohorts comprising the Dutch sample contributed data to the 2022 study cited above.
The R1866W variant is classified as “uncertain: most likely not pathogenic” by the criteria of Holstege et al. (Holstege et al., 2017).
Functional Consequences
The R1866W variant was predicted to be deleterious by SIFT and PolyPhen-2, but neutral by Mutation Taster (Campion et al., 2019).
In a study investigating the effects of SORL1 missense mutations on protein processing, the R1866W variant did not affect the maturation (glycosylation) of SORL1 overexpressed in HEK293 cells (Rovelet-Lecrux et al., 2021).
