Findings
In a European-American cohort, one of 134 late-onset Alzheimer’s cases and one of 266 controls were heterozygous carriers of this variant (odds ratio: 1.989, p = 0.6206) (Fernández et al., 2016).
In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, this allele was observed once among the controls (Holstege et al., 2022).
Functional Consequences
Arginine-2163 is part of a highly conserved sequence (KHRR) in SORL1’s cytoplasmic tail. These four amino acids compose a nuclear localization signal for the cytoplasmic tail, which can be released following TACE-mediated ectodomain shedding and γ-secretase cleavage of the remaining C-terminal fragment (Böhm et al., 2006). The effect of the arginine-to-glutamine substitution is not yet known.
The variant was predicted by PolyPhen-2 to be probably damaging (Fernández et al., 2016).
