Findings

In a European-American cohort of 134 sporadic late-onset Alzheimer’s cases and 266 controls, one control subject was found to be a heterozygous carrier of this variant (Fernández et al., 2016).

Another control carrier was found in a data set from the Alzheimer Disease Exome Sequencing France (ADESFR) project, containing 927 late-onset Alzheimer’s, 852 early-onset cases, and 1273 controls (Campion et al., 2019).

In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, including ADESFR, this allele was observed once among the controls (Holstege et al., 2022).

Functional Consequences

The Y1922S variant was predicted to be tolerated by SIFT and benign by PolyPhen-2, but deleterious by Mutation Taster (Campion et al., 2019).
