I contributed to our understanding of the blood-brain barrier and developed an osmotic method to enhancing drug entry into the brain for treating brain tumors. I used positron emission tomography (PET) to characterize changes in brain blood flow and glucose metabolism in human aging and Alzheimer disease. I also developed methods to quantify and image brain fatty acid metabolism in unanesthetized rodents and humans. With these methods, I elucidate roles of the polyunsaturated arachidonic and docosahexaenoic acids in neurotransmission and neuroinflammation. Recent findings that mood stabilizers selectively downregulate rat brain arachidonic acid metabolism suggest a mechanism for their efficacy in bipolar disorder.
